Green Bioactives Ltd aim to develop plant cell culture as a platform to manufacture safe and sustainable plant biomolecules. We grew plant cells using different types of microbial/mammalian cell culture systems, including a 2.5 L stirred bioreactor and a 20L rocking motion bioreactor. We experienced relatively slow plant cell growth rates and a high risk of culture contamination using these two systems.... we came across the Cellexus CellMaker when we were looking to evaluate airlift bioreactors. During the first run in the CellMaker, our plant cells displayed growth rates comparable to the growth rate achieved using shaking flask liquid culture, with the cells reaching a high density. We are a small company that needed to quickly upscale. We were looking to increase our production for some large-scale trials, so we were looking for a bioreactor that is easy to manage and set up, but we didn’t have autoclaving facilities. The Cellexus CellMaker ticked all these boxes: we got an increased phage yield, it is compact, the simple interface means we could set it up and leave it alone, plus the sterilised disposable bags cuts out any contamination risks. This will be important for us as we move into practices that require more stringent manufacturing processes, such as the medical field. The CellMaker system has allowed us to grow in scale and to achieve higher productivity. Instead of scaling “out” by using several flasks, we can combine it all in one bag inside the bioreactor. In the 8L bioreactor we can easily get between 3 and 8 litres of phage lysate, and in the 50L bioreactor somewhere between 10 and 50 litres in one go, and with higher phage concentration per unit of volume. The CellMaker was the most cost-effective option on the market for a single-use system with temperature control. It is one of the least expensive I have heard about, and this goes for the bioreactor bags as well. When you balance the cost-benefit, the CellMaker is a good option for phage biotechs.
